Biography

Purushottam Narute is postdoctoral research fellow at Critical Care Medicine Department, National Institutes of Health Clinical Center, Bethesda, MD, USA. Purushottam Narute received his BVSc & AH (equivalent of DVM) in 2003 from MAFSU, India. He then completed MVSc in Veterinary Biotechnology at Indian Veterinary Research Institute and worked on antivirals for BoHV-1 as well as vaccine development for Rabies. He then received PhD  in 2012 from University of Pittsburgh, School of Medicine, PA, USA and worked on HIV-1Nef and its interaction with Src family of kinases and discovered compounds that specifically inhibits HIV-1 replication in cell culture.

  • Developed circulatory miRNA profiling of human biofluids
  • Biomarker development for ALI by infectious and non-infectious etiology
  • Developed miRNA profile for immune cells
  • Standardized protocols for an air-liquid culture system for bronchial epithelial cells
  • Demonstrated for first time that Src family kinase activation is a highly conserved property of HIV-1 Nef isolates
  • Successfully completed more than 200 gene cloning
  • Purified more than 30 recombinant proteins using affinity, ion-exchange and size exclusion chromatography on ÄKTAexplorer and BioLogic DuoFlow
  • Assisted in development of X-ray crystallographic structure of Nef-SFK interactions
  • Developed Lentivirus expression system for efficient transduction of suspension and embryonic stem cells
  • Successfully generated Tet-On regulated cell lines
  • Standardized protocols for gene-expression profiling using Real-time qPCR
  • Adapted protocols for HIV-1 replication / inhibition assays
  • Generated chimeric HIV-1 expressing primary Nef isolates for drug screening
  • Experience in handling cell culture, bacteria, virus and yeast
  • Adapted  protocols  for  in  vitro  FRET-based  kinase  screening  assay  for  Nef-mediated  Hck activation
  • Standardized yeast-based kinase assay to evaluate the effect of HIV-1 Nef and HIV-1 Vif on Src family kinases
